Adipose-derived mesenchymal stem cells (ADSCs) are capable of migration to injured tissue sites, where they aid in the repair of the damage. Hepatocyte growth factor (HGF) is critical for damage repair due to its anti-apoptotic, anti-fibrotic and cell regeneration-promoting effects. 赵建辉1,李龙2,杨阳1,易成刚1,刁建升1,夏炜-,刘蓓1,郭树忠1(1.第四军医大学西京医院全军整形外科研究所陕西西安710032;2.第四军医大学口腔医院口腔颌面外科) 【摘要]目的:在体外从脂肪块中分离出脂肪干细胞(adipose derived Stem/stromal cells,ASCs),对其进行形态观察、干细胞鉴定、增殖和分化能力检测。方法:将腹部取皮术的皮下脂肪利用胶原酶消化法,进行体外分离培养,取第3代的细胞进行细胞爬片腿染色、流式鉴定、MTT、细胞周期检测等,利用成脂和成骨培养液诱导,油红O和茜素红染色鉴定诱导结果。结果:原代培养第1次换液时细胞多呈多角形和短梭形,第3代ASCs细胞爬片腿染色显示形态多为长梭形,呈漩涡状生长;流式鉴定显示:CD29+,CD31一,CD34一,CD44+,CD45一,CD49+,CDl06一,CDl33‘;MTT显示ASCs生长增殖活性强;细胞周期检测结果显示:G1=86.8%,G2=8.77%;成脂诱导后油红O染色阳性,对照组为阴性;成骨诱导后茜素红染色阳性,对照组为阴性。结论:人ASCs具有贴壁生长、多向分化以及干细胞表型等特征,且生长增殖活性强,是一种很有应用前景的间充质干细胞。[关键词]脂肪干细胞;培养;鉴定;分化[中图分类号]Q813.1 [文献标识码]A[文章编号]1008-6455(2011)08-1232-06  自从2001年Zuk等“1人从抽脂脂肪中发现脂肪干细胞(adipose derived stem/stromal cells, ASCs)以来,关于ASCs的研究迅速增多,现己成为整形美容外科研究热点之一。ASCs是一群成纤维细胞样的基质干细胞,能分化为不同的细胞系啪,包括脂肪细胞【3】、骨细胞‘“、软骨细胞[5]、骨骼肌细胞[6】、心肌细胞Ⅲ、内皮细胞f8】、神...
